Description of Research Expertise

There is increasing evidence from animal models that activation of innate and adaptive immunity may contribute to the development of epilepsy. In addition, studies of surgical tissue removed from patients with uncontrolled seizures demonstrate persistent inflammation despite treatment with conventional antiepileptic drugs, suggesting that this process may be a target for novel therapies. Our laboratory is investigating the potential role of cytokines and chemokines in modulating neuronal excitability. Specifically, we have found that several pro-inflammatory cytokines are highly upregulated in both rodent and human epileptic brain. Interestingly, we have also found that the expression of these cytokines at baseline vary with age. Certain cytokines peak during adolescence, while others demonstrate elevated levels with advancing age, suggesting specific age windows of vulnerability that may predispose to brain inflammation. Currently, we are assessing the acute and chronic effects of these cytokines on neuronal function in vitro in order to begin to elucidate their mechanisms of action. In parallel, our laboratory uses several epilepsy mouse models to test the efficacy of new therapies that suppress inflammation to improve both the occurrence of seizures and the development of chronic epilepsy.
