Rapid Scan Transient Spectroscopy - Stopped Flow (SF)

In this approach, millisecond mixing of solutions or light activation initiates chemical reactions with kinetics followed by full absorbance or fluorescence UV-Visible spectra resolved every millisecond for up to minutes.  Kinetic resolution of biologically relevant reactions include ligand binding and release, electron transfer or protein conformational changes.  

  • Olis RSM 1000 absorbance spectrometer: Double monochrometer with spinning slit generates full visible and near UV spectrum every millisecond.  Temperature controlled cuvette holder for non-flow measurements. Stopped-flow accessory with 20 mm flow cell; 2-3 ms mixing time resolution. Optional light excitation by xenon flash or Nd:YAG laser at 532, 355 and 266 nm.
  • Olis RSM 1000 fluorimeter: Double monochrometer with spinning slit generates full visible and near UV emission spectrum every millisecond. Temperature controlled cuvette holder for non-flow samples. Stopped-flow accessory with 20 mm flow cell; 2-3 ms mixing time resolution.  Auxiliary mixer for two-stage mixing and stopped-flow.
