HIV-1 p24 and SIV p27 Gag antigen ELISA assay
The Core offers ELISA quantitation of HIV-1 p24 and SIV p27 Gag antigen.
HIV-1 p24 antigen ELISA assay
The Core now offers two different assays to quantify HIV-1 p24 Gag antigen. Our traditional assay provides very accurate endpoint but may require running at several dilutions for samples to be within the standard range, which is approximately 10 pg/ml – 150 pg/ml. The alternative assay has a larger dynamic range (10 pg/ml to 10,000 pg/ml) and is suitable to assess large fold-differences, but may not be ideal to detect smaller differences. The two assays are internally consistent but may not be inter-convertible.
HIV-1 g24 Gag ELISA (Perkin-Elmer) - low range
Range: 10pg/ml – 150pg/ml, ideal for precise accurate end-point quantification
Volume required: 225 per well
Samples: Samples (225 μl) should be submitted in their final dilution desired in a 96 well tissue culture plate. Place plate in a clearly labeled zip lock bag, freeze and then bring to us. Dilutions should be done in tissue culture medium containing 5-10% serum. This assay is run multiple times per week. Charges for HIV-1 p24 (Perkin Elmer) range from $4.00 to $6.00 per sample, based on the number of samples submitted.
HIV-1 p24 Gag Alphalisa (Perkin-Elmer) - high range
Range: 10pg/ml – 10,000pg/ml, ideal for large differences although may be less accurate for small fold-differences
Volume required: 15 μl of sample (undiluted, please contact us prior to discuss dilutions).
Samples: Samples (15 μl) should be provided in a 96 well plate (or tubes for smaller number of samples). The assay is run upon Core users request. Price upon request.
SIV p27 Gag assay
The SIV p27 Gag assay has a dynamic range of 10pg/ml – 2000pg/ml. SIV p27 assays are scheduled on an as-needed basis with users. A sample of 125 μl for p27 ELISA assay is required and should be submitted in their final dilution form in 96 well tissue culture plate (place plate in zip lock bag, freeze and then bring to us). Dilutions must be done in tissue culture medium containing 5-10% serum. The results are provided at the end of the day when assay is performed. For SIV p27 ELISA assay, charges range from $4.75 to $7.50 based on the sample number.
