Transgenomic's (www.transgenomic.com) Wave DHPLC system (denaturing HPLC ) has been up and running in the DNA Sequencing Facility for about two years. So far we have successfully analyzed a number of different genes for mutations and polymorphisms. Also we have used DHPLC for fragment separation. and purification. We have recently upgraded the system to increase the throughput significantly. We have also set up Single Base Extension (SBE) based genotyping of SNP and/or mutations.
For mutation scanning about 30 µl unlabeled PCR products are to be submitted in a PCR plate or 0.2 µl strip tubes., No purification is necessary. Submit the sequence of the PCR products including both primers on a disk or by e-mail. After the run the colored chromatograms are printed and training provided at no extra charge for detection of heterozygotes.
