Penn Vector Core: Services and Rates

The Penn Vector Core has over a decade of experience in viral based vector production and currently offers vectors based on AAV, adenovirus and lentiviruses.
All of the vectors produced in our facility are:
- produced in a state-of-the art facility in the Translational Research Building at Penn
- preclinical grade generated in the spirit of GMP using standard operating procedures
- produced using fully characterized source materials and master seed stocks
- quality controlled by an independent QC Core using a variety of Quality Control assays
- subject to release criteria developed at Penn for preclinical grade vectors
Our core is the only vector core to establish release criteria that must be met by the vectors produced in our facility. If we do not meet the release criteria we have established for our vectors, we will repeat the preparation once again at not additional charge to the investigator. If the production of a vector must be repeated, the investigator will be billed only for the initial production and be provided with the preparation that resulted in the highest yield.
AAV Vector Services: A number of novel AAV vectors were originally isolated and developed at Penn in the laboratory of Dr. James Wilson and first made available to investigators through the Penn Vector Core. Due to its close proximity to the Wilson laboratory, the Penn Vector Core is able to rapidly assimilate new vector technologies and make them available to its users. The Core offers a variety of novel serotype AAV vectors and additional vectors currently under development will be distributed through the Penn Vector.
The Core currently offers 8 AAV vector serotypes, with additional serotypes also available. The tropism of selected AAV serotypes is shown in the following table with an X shown where efficacy has been demonstrated. For investigators wishing to discuss the serotype of choice for their application, please contact the Director of the Penn Vector Core.
For first time users interested in testing a variety of AAV vectors for a particular application, the Core can provide a panel of 6 AAV serotype vectors expressing reporter genes for pilot serotype selection studies. We recommend serotype selection studies be performed in vivo since the in vitro transduction efficiencies of many of the serotypes below does not accurately reflect in vivo performance.)

AAV Vector Release Criteria: The expected yields shown above are based on the average yields of reporter gene vectors generated for AAV serotypes 1, 5, 7, 8, 9 and rh10. (AAV serotypes 2 and 6 are somewhat lower yielding; please contact the vector core for expected yields.) Although we try our best to achieve the same yields for custom vector preparations as we do for our reporter gene vectors, our release criteria for custom vectors is ½ of the anticipated yield of our reporter gene preparations. If we do not meet the release criteria we have established for our vectors, we will repeat the preparation once again at not additional charge to the investigator. If the production of a vector must be repeated, the investigator will be billed only for the initial production and be provided with the preparation that resulted in the highest yield.
Our release criteria do not apply to AAV vectors with the following:
- oversize genomes (>4.9 kb from ITR-ITR including ITRs)
- self-complementary or double-stranded genomes
- genomes expressing shRNA under Pol II or Pol III promoters
- genomes expressing toxic genes under ubiquitous promoters
Vectors for which release criteria do not apply will be produced with no guarantee of yield.

Rates for Support Service:
The rates for support services including cloning (one-step), DNA amplification (for routine scale production) and DNA structure and sequence analysis are provided in the table below. Note that DNA structure and sequence analysis services are required if in support of custom cloning services provided by the core and include full genome (eg. from AAV 5’ ITR to 3’ ITR) sequence analysis as well as thorough restriction enzyme analysis.
