PAS - McMANNUS' Periodic Acid Schiff's

Solution

0.5% Periodic Acid: (prepare fresh)

Periodic Acid 0.5g (Sigma P7875)

ddH₂O 100ml

Schiff's Reagent (Sigma 395-2-016 )

Store at 4°C. Caution: Carcinogen

0.1-0.5% Amylase Solution (Use as a negative control

ddH₂O 100ml ( prewarm at 37 °C for 1 hour )

Amylase 0.1-0.5g (Sigma A6814)

Add amylase before use. Store amylase at-20 °C.

Procedure

1. Deparaffinize and hydrate to distilled water.
2. Add Amylase to prewarmed ddH₂O. Incubate negative control slides in Amylase Solution for 15 min at 37 °C.
3. Rinse in running tap water, rinse in ddH₂O.
4. Place slides into 0.5% Periodic Acid for 5 min.
5. Rinse in ddH₂O.
6. Place slides into Schiff's Reagent for 10-30 min at RT. Watch until tissue turn to deep magenta.
7. Wash in running tap water for 5 min.
8. Counterstain in Harris's hematoxylin for 1 min.
9. Wash in running tap water for 10 min.
10. Dehydrate and mount.
