PicoPure RNA Isolation Kit for recovery of total RNA from less then ten cells could be used for samples containing up to 100 μg of RNA. (Arcturus Bioschience, Inc.)

HistoGene LCM Frozen Section Staining Kit differential staining of nuclei (purple) and cytoplasm (pink). Fast staining protocol. (Arcturus Bioschience, Inc.)

RiboAmp RNA Amplification Kit from as little as 1ng of total cellular RNA; messenger RNA amplified up to 100 folds in one synthesis round and up to 1,000,000 fold in two rounds and produces amplifies antisense RNA (aRNA), ready for labeling and microarray hybridization or QRT-PCR. (Arcturus Bioschience, Inc.)

Turbo Labeling Kits - optimized for labeling of amplified RNA (aRNA) for gene expression profiling. (Molecular Devices).

Paradise Reagent System for formalin fixed, paraffin-embedded tissue samples for gene expression analysis. Helps to establish starting RNA content and integrity in archived tissues for possible downstream analysis.(Arcturus Bioschience, Inc.)

RNase ZAP provides decontamination of instruments, surfaces, glass, plastic, from RNAse and nucleases (AMBION). Alternatives are available.

Prepare PEN membrane (#1440-1000) slides:

Dip or generously spray the slides with pure RNAse ZAP (or other nuclease decontaminating agent). Follow with two washes in RNAse-free water and drying at room temperature. Slides could be store in RNA free box, until use.

Please note, slides #1440-1600 slides already nuclease free. UV treatment in cell culture hood for 30 minutes will increase hydrophility of the PEN-membrane and promote paraffin and cryosections adherance.

Prepare cryosections:

  • Bring OCT mounted tissue in RNAse free cryostat, equilibrate for 5-10 min.
  • Cut sections at 8-10 μm. Make sure to change or wipe knife and surrounding surfaces in between different specimens.
  • Mount sections on room temperature RNAse free slide.
  • Transfer immediately to RNA-se free box, kept on dry ice.
  • Store slides at -80°C. For best results use sections within 0-7 days.
