Quantitative Analysis of Structural Connectome Abnormalities

Many of the clinical and behavioral manifestations of diffuse brain injuries such as Traumatic Brain Injury (TBI) are thought to arise from disruption to the structural network of the brain due to diffuse axonal injury. The heterogeneous nature of such injuries highlights the need for subject-specific analyses that can facilitate development of patient-oriented diagnostic and prognostic tools. Additionally, the diffuse nature of such disorders, with many connections affected to varying extents, necessitates the development of clinically relevant summary measures for assessing the overall injury burden. We develop new measures to quantify disruption of the structural connectome, in order to define subject-specific measures of connectome injury that summarize the cumulative effects of connectivity abnormalities in the entire brain network. One such measure is Disruption Index of the Structural Connectome (DISC).
