Description of Research Expertise

I lead a laboratory whose overall focus is on the mechanisms by which subpopulations of tumor cells within head and neck cancers evade therapy. Our work has pursued how stem cell-like subpopulations with mesenchymal differentiation resist anti-EGFR therapy via both tumor cell-autonomous mechanisms and crosstalk with stromal cells. We have recently sought more precise molecular definition of cancer stem cells within oral cancers as well as detailed understanding of mechanisms involved in regulating their homeostasis. By defining mechanisms responsible for replenishing such cells after their targeted depletion, we are seeking to establish novel approaches for durable cancer stem cell eradication. In the process, we have developed the expertise and infrastructure for analyzing stem cell-like subpopulations within cell lines, patient-derived xenografts, and primary human oral cancer samples based on molecular and functional criteria. We are using lentiviral cDNA and shRNA vectors to investigate the functional crosstalk among genes and signaling pathways involved in cancer stem cell maintenance.
