Whole body energy balance

Promethion metabolic cage system
A 16-cage indirect calorimetry system by Sable Systems, that measures VO2, VCO2, RER, energy expenditure, food intake, water intake, and activity levels in a light/dark and temperature-controlled incubator (4 deg C to 30 deg C), with optional running wheels and food access control doors for pair feeding.

13C Stable isotope gas Analyzer
Enables measurement of 13C exhaled in the breath, used concurrently with the Promethion metabolic cage system for stable isotope infusions or bolus treatments.

Stable isotope infusions
Infusion of stable isotopes through the jugular vein and blood sampling from the carotid artery.

Concurrent infusion, blood sampling, and indirect calorimetry gas analysis of VO2, VCO2, RER, energy expenditure, with optional 13C stable isotope gas analysis and ambient temperature options from 4C to 30C.

Glucose clamp (hyperinsulinemic-euglycemic clamp with [3-3H]glucose and 2-[14C]deoxyglucose)
Clamp procedures in the conscious, un-restrained mouse (includes surgery, analysis and plasma insulin measurement). This procedure will allow investigators to assess whole-body insulin action (GIR) and to distinguish between insulin’s effect on endogenous glucose production (EndoRa), glucose utilization (Rd) and to examine insulin’s effect on specific tissues (Rg; muscle, fat, heart, brain). 

Glucose tolerance test
Intraperitoneal or oral delivery of glucose to assess glucose tolerance. 

Insulin tolerance test
Intraperitoneal injection of insulin to assess insulin tolerance. 

Fasting plasma insulin
Plasma collection then insulin ELISA.

Other Services

Collection of any desired tissue.

Tissue fatty acid uptake is assessed by measuring tissue-specific incorporation of circulating R-2-Bromopalmitic acid [9,10-3H] following intraveous injection which is not metabolized via β-oxidation.

The RMPC works with IDEXX BioAnalytics to measure the analytes listed below in plasma or serum. Please reach out for specific sample collection guidelines if you are interested in this service.
